Arthur Mallay Lesk ist ein Proteinforscher und Professor für Biochemie und Molekularbiologie an der Pennsylvania State University in University Park. Lesk erhielt 1961 einen Bachelor-Abschluss, magna cum laude, von der Harvard University. Er promovierte 1966 an der Princeton University. Außerdem erhielt er 1999 einen Master-Abschluss von der University of Cambridge im Vereinigten Königreich. Lesk hat bedeutende Beiträge geleistet zum Studium der Proteinevolution. Er und Cyrus Chothia, die am Labor für Molekularbiologie des Medical Research Council (UK) in Cambridge, Großbritannien, arbeiten, entdeckten die Beziehung zwischen Änderungen in der Aminosäuresequenz und Änderungen in der Proteinstruktur, indem sie den Evolutionsmechanismus in Proteinfamilien analysierten Entdeckung hat die quantitative Grundlage für die erfolgreichste und am weitesten verbreitete Methode zur Strukturvorhersage, bekannt als Homologiemodellierung, geliefert.
