Peter Straub est né à Milwaukee, Wisconsin, fils de Gordon Anthony Straub et d'Elvena (Nilsestuen) Straub.
Straub a lu avec voracité dès son plus jeune âge, mais ses intérêts littéraires n'ont pas plu à ses parents; son père espérait qu'il deviendrait un athlète professionnel, tandis que sa mère voulait qu'il soit ministre luthérien. Il a fréquenté la Milwaukee Country Day School grâce à une bourse et, pendant son séjour là-bas, a commencé à écrire.
Straub a obtenu un baccalauréat spécialisé en anglais à l'Université du Wisconsin-Madison en 1965 et une maîtrise à l'Université Columbia un an plus tard. Il a brièvement enseigné l'anglais au Milwaukee Country Day, puis a déménagé à Dublin, en Irlande, en 1969 pour travailler sur un doctorat et commencer à écrire professionnellement.
Après un succès mitigé avec deux tentatives de romans littéraires grand public au milieu des années 1970 ("Mariages" et "Sous Vénus"), Straub s'est lancé dans le surnaturel pour la première fois avec "Julia" (1975). Il a ensuite écrit "Si vous pouviez me voir maintenant" (1977) et a attiré l'attention du grand public avec son cinquième roman, "Ghost Story" (1979), qui a été un succès critique et a ensuite été adapté dans un film de 1981. Plusieurs romans d'horreur ont suivi, avec un succès croissant, dont "The Talisman" et "Black House", deux collaborations d'horreur fantastique avec l'ami de longue date de Straub et collègue auteur Stephen King.
En plus de ses nombreux romans, il a publié plusieurs ouvrages de poésie au cours de sa vie.
En 1966, Straub a épousé Susan Bitker. Ils ont eu deux enfants; leur fille, Emma Straub, est également romancière. La famille a vécu à Dublin de 1969 à 1972, à Londres de 1972 à 1979 et dans la région de New York à partir de 1979.
Straub est décédé le 4 septembre 2022, à l'âge de 79 ans, des complications d'une fracture de la hanche. Au moment de sa mort, lui et sa femme vivaient à Brooklyn (New York).
