Peter Straub è nato a Milwaukee, nel Wisconsin, figlio di Gordon Anthony Straub e Elvena (Nilsestuen) Straub.
Straub leggeva voracemente fin dalla tenera età, ma i suoi interessi letterari non piacevano ai suoi genitori; suo padre sperava che sarebbe diventato un atleta professionista, mentre sua madre voleva che fosse un ministro luterano. Ha frequentato la Milwaukee Country Day School con una borsa di studio e, durante la sua permanenza lì, ha iniziato a scrivere.
Straub ha conseguito una laurea con lode in inglese presso l'Università del Wisconsin-Madison nel 1965 e un master alla Columbia University un anno dopo. Insegnò brevemente inglese al Milwaukee Country Day, poi si trasferì a Dublino, in Irlanda, nel 1969 per lavorare su un dottorato di ricerca e iniziare a scrivere professionalmente
Dopo un successo misto con due tentativi di romanzi letterari mainstream a metà degli anni '70 ("I matrimoni" e "Sotto Venere"), Straub si è dilettato nel soprannaturale per la prima volta con "Julia" (1975). Ha poi scritto "If You Could See Me Now" (1977) e ha attirato l'attenzione del pubblico con il suo quinto romanzo, "Ghost Story" (1979), che è stato un successo di critica ed è stato successivamente adattato in un film del 1981. Seguirono diversi romanzi horror, con crescente successo, tra cui "The Talisman" e "Black House", due collaborazioni fantasy-horror con l'amico di lunga data di Straub e collega autore Stephen King.
Oltre ai suoi numerosi romanzi, durante la sua vita pubblicò diverse opere di poesia.
Nel 1966, Straub sposò Susan Bitker. Hanno avuto due figli; anche la loro figlia, Emma Straub, è una scrittrice. La famiglia ha vissuto a Dublino dal 1969 al 1972, a Londra dal 1972 al 1979 e nell'area di New York City dal 1979 in poi.
Straub è morto il 4 settembre 2022, all'età di 79 anni, per complicazioni dovute a una frattura all'anca. Al momento della sua morte, lui e sua moglie vivevano a Brooklyn (New York City).
